Application of Laser Confocal Scanning Microscopy in the Expression of Cx43 in Bone Marrow Stromal Cells of Acute Leukemia
To investigate the application of laser confocal scanning microscopy in the expression of connexin (Cx43) on bone marrow stromal cells of acute leukemia and to understand the functional mechanism of gap junctional communication (GJIC) in bone marrow stromal cells of acute leukemia. The samples of acute leukemia bone marrow stromal cells cultured in vitro in the Department of Hematology of our hospital were selected and included in the experimental group, while the samples of normal bone marrow stromal cells were included in the control group.Cx43 expression in the two groups was detected and compared by laser confocal scanning microscopy, and GJIC function of the two groups was compared according to fluorescence bleaching recovery technology. The quantitative results of Cx43 showed that the pixel density of Cx43 in the experimental group (34.15 ±18.06)% was lower than that in the control group (80.93± 10.11)%, and the difference between the two groups was statistically significant (P < 0.05); The results of Cx43 localization showed that Cx43 was not expressed in the nucleus and mainly expressed in the cytoplasm and membrane. Among them, the experimental group was mainly expressed in the cytoplasm, while the control group was mainly expressed in the cell membrane. The test results of fluorescence bleaching recovery technology show that: The maximum recovery ratio of fluorescence intensity in the experimental group within 400 s was (38.22± 3.05)% lower than that in the control group within 400 s after quenching (84.31± 2.88)%. There was a statistical significance between the two groups (P < 0.05). The average fluorescence recovery rate of the experimental group (5.60± 0.98)%/min was lower than that of the control group (19.18± 1.42)%/min, and the difference between the two groups was statistically significant (P < 0.05). (1) Cx43 quantification and localization can be clearly detected by laser confocal scanning microscopy, and detection by fluorescent bleaching recovery technology is helpful to understand the functional mechanism of GJIC; (2) Cx43 expression in bone marrow stromal cells of acute leukemia is lower than that in normal bone marrow, and there is abnormal localization, and its GJIC function decreases.
